In Vivo and in Vitro Characteristics of Interleukin 6-transfected B16 Melanoma Cells1

چکیده
Interleukin 6 (11-6) is a multifunctional cytokine important in the inflammatory response. Its potential role as an antitumor agent has been suggested by its demonstrated activity in a variety of tumor models. The mechanism of antitumor activity has been proposed to be its en hancement of cytotoxic T-cell function. In the current work we demon strate clear antitumor activity for this cytokine in a nonimmunogenic tumor system. B16 melanoma cells transfected with the human IL-6 complementary DNA demonstrated slower tumor growth in vivo. Tu mors that developed from these cells had a prominent stromal matrix, an easily recognized infiltration of inflammatory cells, fewer mitotic figures, and fewer blood vessels. These in vivo findings corresponded with a greater adhesion of the IL-6-transfected B16 cells to stromal matrix proteins (laminin, fibronectin, and vitronectin) and a less prom inent vascular response in an intradermal angiogenesis assay. There fore, we propose that with weakly antigenic tumors, such as B16 mela noma, IL-6 may mediate important antitumor responses by nonspecific proinflammatory mechanisms.

متن کاملThe role of microRNA-30a and downstream snail1 on the growth and metastasis of melanoma tumor
Objective(s): Growing evidences have indicated microRNAs as modulators of tumor development and aggression. On the other hand, a phenomenon known as epithelial-mesenchymal transition (EMT) that indicates a transient phase from epithelial-like features to mesenchymal phenotype is a key player in tumor progression. In this study, we aimed to assess the potential impacts...
متن کاملThe antitumor activity of a doxorubicin loaded, iRGD-modified sterically-stabilized liposome on B16-F10 melanoma cells: in vitro and in vivo evaluation
Considering the fact that iRGD (tumor-homing peptide) demonstrates tumor-targeting and tumor-penetrating activity, and that B16-F10 (murine melanoma) cells overexpress both αv integrin receptor and neuropilin-1 (NRP-1), the purpose of this study was to prepare a novel doxorubicin (DOX)-loaded, iRGD-modified, sterically-stabilized liposome (SSL) (iRGD-SSL-DOX) in order to evaluate its antitumor ...
متن کاملT cell-mediated, IFN-gamma-facilitated rejection of murine B16 melanomas.
The murine melanoma cell line B16.F10 (H-2b) was used to study specific T cell responses that reject tumors. Stable B16 transfectants were established that express viral Ags, either the hepatitis B surface Ag (HBsAg) or the large tumor Ag (T-Ag) of SV40. B16 cells and their transfected sublines were CD40+ CD44+ but expressed no (or low levels of the) costimulator molecules CD154 (CD40L), CD48, ...
متن کاملThe effect of low intensity dual frequency ultrasonic waves on the viability of the B16-F10 melanoma cell
In this study, the effect of single and dual-frequency sonication on cell death of B16-F10 melanoma cells is investigated at constant temperature. Here, 20 groups were studied. The test groups consisted of: control and sham, 40 kHz (intensity: 0.24 W/cm2), 1 MHZ (intensity: 0.5 W/cm2) and the dual frequency groups which each frequency group included seven subgroups of 30, 120, 60, 150, 300, 600...
